The final steps in the initiation of DNA replication: Activation of the pre-replication complex

Precise replication of genomic DNA is essential for the propagation of all life. Unlike bacteria, which usually initiate replication from a single origin per chromosome, eukaryotes initiate DNA replication from hundreds or thousands of sites. Sequence designation of these sites is not yet fully understood, but the many proteins that orchestrate the timing and location of initiation have been extensively studied. Genes encoding most of these have been found ubiquitously in fungal and metazoan genomes. Taking advantage of yeast genetics, I have studied the molecules that regulate timing of the initiation of DNA replication in Saccharomyces cerevisiae
